The Wimbledon champion fell to Clara Tauson of Denmark with a score of 7-6 (7-1) 6-3.
Swiatek joins top seed Coco Gauff, two-time defending champion Jessica Pegula, and fourth seed Mirra Andreeva, who were also knocked out earlier in the tournament.
In other matches, Naomi Osaka delivered a dominant performance, winning 6-1 6-0 against Anastasija Sevastova.
In the men’s event, Australia’s Alex de Minaur advanced to the quarter-finals by defeating Frances Tiafoe, while fifth seed Holger Rune was upset by Alexei Popyrin.
